1 范围
本文件描述了饲用微生物制剂中产朊假丝酵母(Candida utilis)菌数的测定方法。
本文件适用于含有产朊假丝酵母饲用微生物制剂的测定。

3 术语和定义

4 试剂或材料
除非另有规定,仅使用分析纯试剂。
4.1 水:GB/T 6682,三级。
4.2 0.85% 灭菌生理盐水:准确称取8.5 g 氯化钠加入1 000 mL 水中,搅拌至完全溶解,分装后,121 ℃
灭菌30 min,备用。
4.3 孟加拉红琼脂培养基:见附录A 中A.1。
4.4 酵母膏胨葡萄糖(YPD)琼脂培养基:见A.2。
5 仪器和设备
除微生物实验室常规灭菌及培养设备外,其他仪器设备如下。
5.1 培养箱:可控精度 ±1 ℃。
5.2 天平:感量为0.1 g。
5.3 光学显微镜:400 倍~1 000 倍。
5.4 拍击式均质器及无菌均质袋。
5.5 旋涡混合器。
5.6 无菌培养皿:直径90 mm。
5.7 聚合酶链式反应(PCR)仪。
6 采样
按照GB/T 42959 执行。

7 试验步骤
7.1 样品稀释
7.1.1 固体、半固体或液体样品:称取25 g 样品或以无菌吸管吸取25 mL 样品,置于装有225 mL
0.85% 灭菌生理盐水(4.2)的500 mL 无菌锥形瓶(瓶内预置适当数量的无菌玻璃珠)中或无菌均质袋
(5.4)中,充分振摇或用拍击式均质器拍打1 min~2 min,制成1∶10 样品匀液。
7.1.2 取1 mL 1∶10 样品匀液注入含有9 mL 0.85% 灭菌生理盐水(4.2)的试管中,在旋涡混合器
(5.5)中混匀,此液为1∶1
